I had to drill out the two screws that old the rear hub on and I inadvertently drilled too wide smoothing out the threads.
How can I re-drill threads or tap new holes. Never done that before. Is there a special tool involved? Is it difficult?
Or, should I just drop $128 for a new hub?

To be honest,I dont see a really big need for those 2 little screws in the rear hub. After all, the wheel will hold the drum on once the lugnuts are tightened down. Or you might be able to over drill and retap to the next larger size.
Are you speaking of the short counter sunk slotted head screws that hold the drum to the axle flange? If so don't sweat it and just move on. They are not necessary.

I was a bit confused at first read of the post but it must be the drum screws Its the only thing back there that comes in pairs.

It is the two screws that hold the Drum to the axle. And yes, they are recessed.
Good. I suppose the lugs really do most of the "holding" anyway.
I went to the trouble of locating and replacing those screws; For the life of me I don't know why.
I had a customer who had them back ordered just for a matter of honor. They are good for holding things together when you are moving a rear end about out of the car.
To answer your original question, yes, you can tap them slightly larger and it's not dificult if you have the right tools. You'd have to drill the drum, counter sink it and find new screws. Since they really don't do anything, why go to the bother?
Ditto, they aren't necessary at all. However, if you really feel the need to have them you could Helicoil the holes or install threaded inserts like Keenserts, etc. I'd spend the time and money elsewhere though.
If you really want to.... turn the drum 90 degrees and drill new holes and tap them..
There are drill bits that are reverse threaded; possibly if you used one of these to drill a pilot hole, you would find that the vibration and heat made the broken part walk out on its' own. HTH!
